Mutational sources of trans-regulatory variation affecting gene expression in Saccharomyces cerevisiae

چکیده
Heritable variation in a gene’s expression arises from mutations impacting cis - and trans -acting components of its regulatory network. Here, we investigate how -regulatory are distributed within the genome gene network by identifying characterizing 69 with effects on same focal Saccharomyces cerevisiae . Relative to 1766 without this gene, found that these were enriched coding sequences transcription factors previously predicted regulate gene. However, over 90% identified mapped other types genes involved diverse biological processes including chromatin state, metabolism, signal transduction. These data show genetic changes can impact , revealing properties provide raw material for segregating natural populations.

CLONING AND EXPRESSION OF HUMAN IFNα2B GENE IN SACCHAROMYCES CEREVISIAE
Interferon is a protein secreted by eucaryotic cells following stimulation by viruses, bacteria, and many other immunogenes. Recent medical studies indicate that interferons have effective role in the treatment of virus infections, immunodeficiency and certain types of cancer such as hairy cell leukaemia (HCL). The aim of the present study is to apply yeast strain for secreting human IFNα2b fol...
متن کاملMutational hypersensitivity of a gene regulatory protein: Saccharomyces cerevisiae Gal80p.
The inhibitor of galactose catabolic (GAL) gene expression in Saccharomyces cerevisiae, Gal80p, interacts with the activator Gal4p and the signal transducer Gal3p and self-associates. Selection for loss of Gal80p inhibitor function yielded gal80 mutants at an extremely high rate. Out of these, 21 nonoverlapping point mutants were identified; each were due to a single-amino-acid exchange in cons...
متن کاملLocal Regulatory Variation in Saccharomyces cerevisiae
Naturally occurring sequence variation that affects gene expression is an important source of phenotypic differences among individuals within a species. We and others have previously shown that such regulatory variation can occur both at the same locus as the gene whose expression it affects (local regulatory variation) and elsewhere in the genome at trans-acting factors. Here we present a deta...
متن کاملCorrelating Gene Expression Variation with cis-Regulatory Polymorphism in Saccharomyces cerevisiae
Identifying the nucleotides that cause gene expression variation is a critical step in dissecting the genetic basis of complex traits. Here, we focus on polymorphisms that are predicted to alter transcription factor binding sites (TFBSs) in the yeast, Saccharomyces cerevisiae. We assembled a confident set of transcription factor motifs using recent protein binding microarray and ChIP-chip data ...
متن کاملIsolation, Subtype Determination, Cloning and Expression of HBsAg Gene from an Iranian Carrier in Saccharomyces cerevisiae
The Hepatitis B Surface antigen ( HBsAg) gene was isolated from an Iranian HBeAg positive carrier by PCR. The gene was cloned in pUC19 for sequencing and pYES2 for expression in Saccharomyces cerevisiae, which pNF1 and pDF3 constructs were made respectively. The sequencing data showed that the isolated HBsAg gene shared more than 90% homology with the ayw subtype. The pDF3 was transferred into ...
